How to dissolve the peptide in powder form?

Peptide in powder form should be dissolved in autoclaved distilled water

 

How to extract DNA from hair follicles?

For hair follicle, GeneAll Blood SV can be used. Here is mini protocol.

Isolation of genomic DNA from hair follicles.

Put 5 ~ 10 hair follicles into a 1.5ml microcentrifuge tube, and add 200 ul Buffer UM01 and 20 ul Proteinase K. Incubate at 56C for at least 1 hour until the sample is dissolved. Invert the tube occasionally to disperse the sample, or place on a rocking platform.

» Buffer UM01(User-made) ; 10mM TrisCl, pH8.0, 10mM EDTA, 100mM NaCl, 40mM DTT, 2% SDS

» Add DTT immediately before use.

  1. Add 200 ul of Buffer BL to the lysate and vortex to mix.
  2. Add 200 ul of Ethanol to the sample and mix by vortexing.
  3. Continue from step 6 at page 15 in the protocol handbook.
  4. Since the cell numbers of the sample(hair) is relatively small, it is recommended the volume of elution should be reduced to 50 ul.


Can DNA from human whole blood which has frozen for about 2-3 years be extracted from GeneAll blood kit? Will the yield be affected? Any additional step shall be taken to maximize the yield?

GeneAll blood kit can be used for the preparation of DNA from the whole blood frozen for 2-3 years. The yields may be decreased depending on the condition of freezing method. If the whole blood had been stored under -70 c and the appropriate conditions, the yields will not be decreased significantly. However, the sample repeated freezing and thawing may give a poor result.

For maximizing the yield, frozen blood should be thawing without any blood clot.

Which buffer (PCR 10X reaction buffer – provided in the kit or buffer and MgCl provided in the Taq) should be used in the reaction?

The researcher should use the buffer coming with the kit.

 

Does researcher need to add MgCl into the reaction if he uses the PCR buffer provided in the kit?

No.

What are the storage for a peptide?

If the peptide has been dissolved in distilled water, it should be stored n -78ºC but only for few weeks. If the peptide is in powder form, it can be stored in -20ºC for 1 year.

 

How to dissolve the peptide in powder form?

Peptide in powder form should be dissolved in autoclaved distilled water.

What is the estimated quantity for 1 OD primer?

Estimated 1 OD = 5 nmols = 30 ug, for a 20 mer oligos.

 

What is the form of supplied primer? And how to dilute it?

The primer is already dissolved in de-ionized water. It can be diluted by using de-ionised water or distilled water (autoclaved). 

 

What is the storage condition for primer and probe?

Although they are stable in room temperature for up to 2 weeks, they should be stored at -20C. Repetitive freeze-thaw cycles should be avoided by storing as aliquots. Storing at concentration above 20 uM is recommended. Oligonucleotids coupled to fluorophores should be stored protected from light. Stored under the above condition.
